چکیده

SUMMARY Tumor-specific transplantation antigen (TSTA) was solu-bilized from cell membranes of sarcoma Meth-A with non-ionic detergent Nonidet P40. Soluble TSTA was partially characterized by Chromatographie separation and electro-phoresis. The antigen responsible for tumor rejection activ ity had a molecular weight of approximately 70,000 daltons in the presence of detergent and an electrophoretic mobility of a-globulin. TSTA was well separated from mouse histo-compatibility antigen H-2 by lectin affinity chromatography. The antigen purified by a sequence of procedures, includ ing gel filtration, lectin affinity chromatography, column electrophoresis, and rechromatography on agarose, showed only three major bands on polyacrylamide gel elec trophoresis. TSTA was specific for sarcoma Meth-A.

Immunochemical evidence of a tumor-specific surface antigen obtained by detergent solubilization of the membranes of a chemically induced sarcoma, meth-A.

We reported previously the partial purification of detergent-solubilized specific tumor rejection antigen of a chemically induced sarcoma, Meth-A. During the course of the study, rabbit antiserum against a partially purified specific tumor rejection antigen preparation was raised and rendered specific by in vivo absorption. In this report we show that an antigenic molecule defined by in vivo-ab...

Purification and biochemical properties of tumor-associated transplantation antigens from methylcholanthrene-induced murine sarcomas.

A tumor-associated transplantation antigen with an apparent molecular weight of 75,000 has been isolated from the cytosol of the BALB/c methylcholanthrene-induced sarcoma, Meth A. The antigen was purified either by preparative electrophoresis in the presence of NaDodSO4 or by immunoaffinity chromatography after hexylamine agarose chromatography, gel filtration, and hydroxylapatite chromatograph...

Extraction of a Murine Tumor-specific Antigen from Cells and Plasma Membranes Using 1-Butano!: Augmentation of Antigen Yield by Colchicine1

The purpose of this investigation was to examine the ability of single-phase aqueous solutions of 1-butano! to release immunoprotective tumor antigen activity from partially purified plasma membranes of the methylcholanthrene-induced fibrosarcoma, MCA-F. Tumor antigen activity was assessed by s.c. immuniza tion of syngeneic C3H/HeJ mice 10 days prior to supralethal challenge. Although brief inc...
